when you need a min of G-U pairs does it make a difference where they are placed when there are multiple locations available and it doesn’t affect the kcal?

I assume you ask for placement of GU’s in challenge and player puzzles. Yes, it do make a difference where they are placed. You should preferable make them line up, that way you can get away with putting more in. You can also put one at the end of a stack. Check the section with stacking tricks in Tips and tricks guide.
