First two CASP 17 targets are Eterna designs!

Holy Cow!!! The first two CASP 17 RNA targets are molecules designed by Eterna players during the OpenKnot rounds! Congratulations everyone! This means the molecules were resolved using cryo-EM and will eventually be deposted in the Protein Database (PDB). The first one is JR. Who designed the second?

It looks like possibly spvincent or a tweak on an spvincent design. Eterna The sequence seems to match GAGCGUAGCACACACUCAGACUAAGUCUGACACACACCUACGCUAAACAUCUUAGUGUGUGAUCGAGAUAUCGAUGUGUGUGUAAGAUGAGAGCGUAGCACACACUCAGACUAAGUCUGACACACACCUACGCUAAACAUCUUAGUGUGUGAUCGAGAUAUCGAUGUGUGUGUAAGAUGA

The first target is one of JR’s entries to the round 4 week 10 puzzle UC2199, which used the secondary structure from one of Ucad’s round 1 designs as determined by SHAPE-based prediction. (JR’s round 4 sequence scored 99.) Interestingly, Ucad’s UC2199 originally was scored as 93 in round 1, but currently shows a score of 81 after the “improved” rescoring. The SHAPE data is pretty clean, so I don’t know why the score dropped so much.

The second target is spvincent’s pilot round design f39. As Jeff points out, the sequences match exactly. The additional characters in the title probably are a notation added by the researcher who performed the cryo-EM. Our SHAPE-based prediction is nice and compact. Looking forward to seeing how it actually folded in the test tube!
